For structural analysis, these complexes were cross-linked with the DSBU cross-linker and analyzed by mass spectrometry. Additionally, the recombinant protein complexes were used as bait proteins to identify interaction partners in nuclear extract.; In dieser Arbeit wurden alle bekannten und essentiellen Proteinkomplexe der mRNA-3‘-Prozessierung (CFI, CFII, CstF, mPSF und der Endonukleasekomplex) rekombinant exprimiert und chromatografisch gereinigt. Zur strukturellen Analyse wurden diese Komplexe mit dem Cross-Linker DSBU chemisch vernetzt und massenspektrometrisch analysiert. Zusätzlich wurden die rekombinanten Proteinkomplexe als Köderprotein eingesetzt, um Interaktionspartner in Kernextrakt zu identifizieren.
